In my home county, we have 1 EMS station that provides coverage for the whole county. Of course, in-game, ambulance only missions are only going to generate close to the EMS station. However, I am wanting ambulance only mission throughout the entire county. How can this be made possible?

If you pay for ambulance extensions at fire halls but don’t put ambulances in them I believe ambulance calls should spawn in that location.

I believe if you send vehicles on patrol calls will spawn around then? So you could set patrol routes around your country for vehicles and then calls will spawn near them.

That is incorrect. This functionality has not been added.
